Cell free Expression systems

The analysis of protein is done through cell free protein expression system for more than decades. Low yield of target proteins was the biggest setback of the cell free systems. Studied through the autoradiography of proteins and Western blotting with the help f some specific antibodies were the mostly used wheat germ or reticulocyte lysates. The value of these tools that are commercialized by various companies represents the fact that the cell free expression system is a valuable method of protein analysis.

Advantages



Cell-free expression systems are powerful and have a few edges over the cell based systems, including PCR linear expressions. The first advantage would be the toxic protein expression, the isotopic protein labelling, the addition of un-natural amino acids and isotic labels, expressing the combined proteins in single reaction, producing small number of proteins rapidly, and addition of cofactors, detergents etc.

A cell free system should be considered as a complementary tool for the expression of protein and its strength should be focused.

Cell free expression reactions and its variants

Cell free reactions are performed in two ways. The reaction vessel that contains all the components that is due to the translation that is being carried out in the batch reaction. The batch system reaction reaches plateau after 2 hours due to several reasons like, diminution of supply of energy, demotion of components such as decreasing concentrations of some ions and nucleotides. With the help of customized batch system the cell free expression system can reach up to a decent level.
